Metabolites found in animals are not present in humans, which could mean that the toxicity observed in that animal species and that can be attributed to metabolites, may not be related to humans. Conversely, a drug metabolite identified during clinical development that is absent in animal test species or at levels in animals that are much lower than in humans, which may indicate the safety testing of drug metabolites need to finish in animals. In such cases, there are two general approaches in safety testing of drug metabolites.
- The first approach is to identify animal species that are commonly used in safety testing. The species forms the metabolites at adequate exposure levels (equivalent to or greater than the human exposure), and then the safety of drug metabolites in this animal species is studied.
