Flavonoids are secondary metabolites that are very abundant in plants, fruits, and seeds. They are responsible for the color, fragrance, and flavor characteristics. They comprise several thousand compounds that possess a common C6-C3-C6 skeleton where two aromatic rings (A and B) are linked through a three-carbon bridge that usually, but not always, forms a heterocyclic ring (C) giving rise to a phenylchromane, which in some cases can be opened giving rise to chalcone and dihydrochalcone forms. On the basis of the substitution pattern of ring C, different flavonoids are distinguished, such as flavones, flavonols, flavanones, flavanols, anthocyanins, dihydroflavonols, and isoflavones. In plants, flavonoids perform many functions like regulating cell growth, attracting pollinators insects, and protecting against biotic and abiotic stresses[1]. In humans, these compounds are associated with a large range of health benefits, such as anti-inflammatory, anticancer, cardio-protective, neuroprotective, antibacterial, antidiabetic, antiparasitic, and antiviral properties. Sample pretreatment
- Solid samples usually need to be homogenized and then dried or frozen in liquid nitrogen.
- Liquid samples are usually first filtered and/or centrifuged, after which the sample is either directly injected into the separation system, or more commonly, the analyte is separated by liquid-liquid extraction or solid-phase extraction.
Separation and detection
- The most commonly used method for separation and detection is column liquid chromatography. Column liquid chromatography of flavonoids is usually carried out in reversed-phase mode, on C8- or C18-bonded silica columns. Gas chromatography, capillary electrophoresis, and thin-layer chromatography are used less frequently than column liquid chromatography.
Identification and structural characterization
- Liquid chromatography-mass chromatography (LC-MS) and on-line liquid chromatography-nuclear magnetic resonance spectroscopy (LC-NMR) are important techniques for the identification of target flavonoids and the structural characterization of unknown compounds.
